An API manufacturing business makes active pharmaceutical ingredients (APIs), the part of a medicine that does the work, such as ibuprofen powder rather than the tablet. It sells them by the kilogram or tonne to companies that turn them into tablets, capsules and injections. The first decision is scale and route. A small multi-product chemical synthesis plant is the realistic entry point: the NPCS project report for a unit making cephalexin, ampicillin and ibuprofen at 500 kg a day each puts total capital investment at ₹958 lakh. Fermentation-based antibiotic projects run into hundreds of crores. Either way, buyers audit your plant and test batches before the first order, so name customers for one or two products before buying land.
Which API should a first plant make: synthesis, fermentation or intermediates?
| Route | What it involves | Capital pointer |
|---|---|---|
| Chemical synthesis, several products | Batch reactions, one product at a time | ₹958 lakh for three APIs (NPCS report) |
| Fermentation-based antibiotics | Fermenters, long scale-up | Clavulanic acid, Nalagarh: ₹450 crore. Penicillin G, Kakinada: ₹1,910 crore (Department of Pharmaceuticals explainer) |
| Intermediates or contract work | Supply intermediates, or make a named API for another company | Depends on the product |
Start with synthesis unless you already have fermentation experience and a funding partner. A multi-product plant protects you: when one API's price falls, the reactors switch to another. Many promoters begin with intermediates; the contract manufacturing route explains how. If this capital is out of reach, the pharmaceutical manufacturing ideas page lists other routes.
How is an API made, and what machinery does the plant need?
A synthesis plant works in batches, and every product follows the same steps.
- Test raw materials, reagents and solvents on arrival.
- Run the reaction in a jacketed stainless steel or glass-lined reactor.
- Separate the product from solvent and by-products; distillation recovers the solvent.
- Crystallise the product by cooling, changing the solvent or adjusting pH.
- Separate and wash the crystals in a centrifuge or filter-dryer.
- Dry the solid, because buyers set limits on residual solvent.
- Mill and sift to the buyer's particle size, then blend into one uniform batch.
- Test against the pharmacopoeia and buyer specification, then pack and release.
The good manufacturing practice (GMP) guideline for APIs, ICH Q7, asks you to declare the step where API manufacture begins. GMP applies from there, and control tightens towards purification and packaging.
| Machine | Job it does |
|---|---|
| Reactors with condensers and receivers | Run the reaction, recover solvent |
| Centrifuge or agitated nutsche filter-dryer | Separate and wash the solid; the filter-dryer also dries it |
| Vacuum tray, fluid bed or rotary dryer | Remove residual solvent |
| Mill, sifter and blender | Control particle size and uniformity |
| Effluent treatment, boiler, chilled water, vacuum, nitrogen | Utilities the plant depends on |
| Quality control lab with HPLC and GC | Test purity and solvents, release batches |
What approvals does an API plant need before the first sale?
| Step | Who decides | What it gets you |
|---|---|---|
| Environmental clearance | Central or state appraisal committee (EIA Notification 2006, item 5(f) for bulk drugs) | Permission to build |
| Manufacturing licence | State drug controller | Form 24 application, Form 25 licence after inspection, naming your technical staff |
| Customer audit | Each buyer | Purchase orders |
| US Drug Master File | You file with USFDA | US customers can cite your process unseen |
| Certificate of Suitability (CEP) | EDQM, Europe | Confirms the API meets the European Pharmacopoeia monograph |
Settle the plot before the clearance route: a plant inside a notified industrial area, or a small unit, falls in the state-level category, and one outside such an area generally goes to the central category. Confirm the form series with your state authority. A Drug Master File is not required by US law, and per FDA's drug master file page it is reviewed only when a customer's application refers to it. A CEP is not a GMP certificate, so inspectors still check the plant.
What does an API plant cost, and what return does the NPCS report show?
The NPCS project report on cephalexin monohydrate, ampicillin trihydrate and ibuprofen lists these figures:
| Item | Figure in the report |
|---|---|
| Capacity | 500 kg a day each of the three APIs |
| Plant and machinery | ₹448 lakh |
| Total capital investment | ₹958 lakh |
| Projected return | 46% |
| Break-even point | 44% |
Plant and machinery is under half the total, and working capital is not shown separately, so get a full costing before you borrow. Costs move with how many products share the reactors and how much solvent recovery and effluent treatment the chemistry needs. Lenders will want a project-specific detailed project report with machinery quotations and buyer letters of intent.
Who buys APIs in India, and how crowded is the field?
India has about 500 API manufacturers, nearly 8% of the global API industry, according to a government factsheet reported by IANS in March 2026. Imports still matter: figures given to Parliament in July 2025 put the import share of APIs by value at about 68% in FY20 and nearly 72% in FY24 (Business Today).
USP's Medicine Supply Map, reported by RAPS in November 2024, shows the competition. India made 50% of new API drug master file filings in 2023 against 32% for China, and held 48% of active filings against 16%. China's new filings rose from 134 in 2021 to 219 in 2023. Filings are not output, but they show where new suppliers enter. Your customers are formulation companies, exporters and other API makers.
Which incentives and bulk drug parks help a new API plant?
Both central schemes turn on what you make and where, not on plant size. The production linked incentive (PLI) scheme for bulk drugs, with ₹6,940 crore to FY 2029-30, covers 41 bulk drugs; by December 2024, 48 projects were selected and 34 commissioned, per the same explainer. Later rounds opened only for named products, such as meropenem and ritonavir in the round that closed on 26 December 2025, and India Briefing reports that chemical synthesis incentives run only to FY 2027-28. Check the Department's PLI portal for open rounds, and see the wider Atmanirbhar Bharat schemes page.
The bulk drug parks scheme (₹3,000 crore) gives up to ₹1,000 crore per park, or 70% of project cost (90% for hilly and north-eastern states), for shared infrastructure such as effluent treatment.
| Park | State | Status as reported |
|---|---|---|
| Jambusar, Bharuch | Gujarat | 800 acres; completion targeted for the first half of 2026 |
| Haroli, Una | Himachal Pradesh | Environmental clearance granted; construction gaining momentum (Rajya Sabha reply, 7 August 2026) |
| Nakkapalli, Anakapalli | Andhra Pradesh | Moved from Kakinada; common infrastructure work in progress as of August 2024 |
Confirm allotment terms with the state agency first.
What goes wrong in a first API plant?
- Qualification outlasts construction. Buyers audit and test before ordering, so working capital must cover a stretch without sales.
- One-product dependence. A price fall leaves the reactors idle.
- Solvent and effluent handling. The chemistry sets the treatment cost, which is easy to under-budget.
- Imported starting materials. Confirm a second source before fixing your costing.
What should you do in the first 30 days?
- Choose one or two APIs and name two buyers for each.
- Check each product's patent position with an intellectual property lawyer.
- Shortlist plots in an industrial area or bulk drug park and ask which clearance category applies.
- Collect quotations for reactors, centrifuge, dryers and effluent treatment.
NPCS has prepared project reports since 1994, more than 5,000 across over 40 sectors. A report for an API plant covers the process with flow sheet, machinery with suppliers, raw materials, manpower, project cost, working capital, cost of production, break-even and a 5-10 year cash flow, in the format banks usually ask for. You can use everything above without it; a lender will want a project-specific report.
Frequently asked questions about API manufacturing
Can you give a list of APIs in common medicines?
Yes. Paracetamol, ibuprofen, cephalexin, ampicillin, atorvastatin, telmisartan and olmesartan are all made in India, and Penicillin G and clavulanic acid are antibiotic ingredients made by fermentation.
Where can I buy active pharmaceutical ingredients?
Buy from licensed manufacturers or their authorised distributors. Ask for the manufacturing licence, a certificate of analysis for each batch, and the DMF or CEP reference if you export.
What is the size of the Indian API market?
No single official figure appears in the sources reviewed, and research firms' estimates differ. The March 2026 government factsheet gives structure instead: about 500 manufacturers and nearly 8% of the global API industry. Name the firm and year if you quote a size.
Who are the top API manufacturers in India?
There is no official ranking. Integrated pharmaceutical groups, API specialists and contract manufacturers all compete, and the order changes with volume, value or filings. Cadila Pharmaceuticals, for example, lists 38 APIs and intermediates with 83 drug master files.
Do I need a Drug Master File to start an API business?
No. FDA says drug master files are not required by statute or regulation and are neither approved nor disapproved. You need one when US-bound customers want to cite your process.
Not part of the page. Everything below this line is for the editor and is not published.
