What is a active pharma ingredients api project report?
An active pharma ingredients API project report is a detailed project report (DPR) for a plant that makes the active substance of a medicine by chemical synthesis. It sets out the molecules, synthesis steps, reactors, solvents, approvals, project cost, means of finance, five-year projections, DSCR and break-even that a lender or investor checks.
Promoters use it to size a bulk drug unit and raise a term loan; banks, investors and approval authorities use it to judge whether the plant can run and repay.
Also known as: API manufacturing project report, bulk drug project report, DPR for API plant, API manufacturing business plan.
Active pharma ingredients API manufacturing plant cost in India
Our sample report models a multi-product API unit with 250 kg per day of capacity, running 330 days a year. Output starts at 60% of capacity and reaches 80% in Year 5, sold at a blended ₹1,930 per kg across the molecules made.
Plant and machinery is just over half of the project cost. Reactors, solvent recovery and the clean room take most of it, so the choice of molecules decides the budget more than the floor area does.

Active pharma ingredients API manufacturing process
Most small API units make several molecules in campaigns on the same reactor train. Each molecule has its own route, but the plant runs every batch through the same eight stages.
- Testing and dispensing
Key starting materials and solvents are tested against specification, then weighed for the batch under a batch manufacturing record.
- Reaction
Each synthesis step runs in a glass lined or stainless steel reactor, with steam or chilled brine in the jacket to hold the set temperature.
- Work-up and isolation
The reaction is quenched, the layers are separated and the solvent is distilled off to leave the crude product.
- Purification
Crude API is treated with activated carbon and recrystallised until related substances fall within the pharmacopoeia limit.
- Centrifuging and drying
A centrifuge separates the crystals, and vacuum tray or fluid bed dryers bring moisture and residual solvents down to specification.
- Milling and blending
Dry API is milled, sifted and blended in a clean room to the particle size the formulation buyer asks for.
- QC release and packing
Each batch is tested by HPLC for assay and impurities and by GC for residual solvents, then packed in double polythene bags inside HDPE drums.
- Solvent recovery
Spent solvent is distilled, tested and reused; the residue goes to the effluent plant or to authorised disposal.
Choosing the molecules and the synthesis route
An API project starts with the molecule list, not the machinery. A first unit usually picks two or three off-patent drugs that share reaction types, so one reactor train can switch between them with a cleaning validation in between.
Most small plants use chemical synthesis. Fermentation-based APIs such as penicillin G and clavulanic acid need large fermenters and much higher investment; the PLI projects for these two products alone cost ₹1,910 crore and ₹450 crore.
For each candidate molecule, the report should test the number of steps, yield per step, solvents used and how much of each can be recovered. A route with one extra step can add a day to the batch cycle and a new effluent stream.
Why the route matters to a lender: the yield and solvent recovery you assume decide raw material cost. A route proven at pilot scale, with batch records, makes the projections credible.
Raw materials for API manufacturing
Key starting materials (KSMs) and intermediates are the largest cost. India still imports many of them, so a report should name at least two suppliers per KSM and price in some stock. In our sample, raw materials cost ₹516.35 lakh in Year 3, about 46.3% of sales.
| Raw material | Share of cost | Year 3 (₹ lakh) |
|---|---|---|
| Key starting materials and intermediates | 58% | 299.48 |
| Solvents (make-up after recovery) | 20% | 103.27 |
| Reagents, acids and bases | 14% | 72.29 |
| Catalysts, activated carbon and filter aids | 8% | 41.31 |
| Total | 100% | 516.35 |
Solvent cost is shown net of recovery. Packing drums and liners are counted separately in the sample.
Machinery for an API plant
Reactors are the largest item in our sample. Their volume and lining set how many batches, and which chemistries, the plant can run.
| Equipment | What it does | Sample cost (₹ lakh) |
|---|---|---|
| Glass lined and SS reactors with condensers | Run the synthesis steps | 94.93 |
| Centrifuges and nutsche filters | Separate crystals from liquor | 40.69 |
| Vacuum tray and fluid bed dryers | Remove moisture and solvent | 33.90 |
| Solvent recovery distillation | Recovers solvent for reuse | 40.69 |
| Clean room powder processing with HVAC | Mills, sifts and blends the final API | 40.68 |
| QC laboratory with HPLC and GC | Tests assay, impurities and residual solvents | 33.90 |
| Boiler, chilling plant and vacuum pumps | Steam, chilled brine and vacuum | 33.90 |
| Effluent treatment with evaporator | Treats process effluent and recovers salts | 20.34 |
| Total plant and machinery | 339.03 | |

Utilities and manpower for an API unit
An API plant runs on steam, chilled brine and vacuum as much as on power. In our sample, power and fuel cost ₹56.44 lakh in Year 3, or 5.1% of sales.
| Steam | Boiler steam for reactor jackets, distillation and dryers |
|---|---|
| Cooling | Cooling tower water for condensers; chilled brine for low-temperature reactions and crystallisation |
| Vacuum and nitrogen | Vacuum pumps for distillation and drying; nitrogen blanketing for reactors handling flammable solvents |
| Effluent | Process effluent goes to an evaporator; state authorities may ask how the unit will reach zero liquid discharge |
| Manpower | 30 people in the sample, costing ₹68.40 lakh in Year 1, including QC chemists and shift operators |
Licences and approvals for API manufacturing
Start the approvals early. The environmental clearance involves appraisal by an expert committee and the drug licence needs an inspection, so plan both alongside the civil work.
| Drug manufacturing licence | Application in Form 24 for a licence in Form 25 from the State Licensing Authority, for drugs outside Schedules C, C(1) and X, after inspection of the premises |
|---|---|
| Good manufacturing practice | Revised Schedule M, notified by G.S.R. 922(E) on 28 December 2023, applies to premises, plant and equipment |
| Environmental clearance | Item 5(f), synthetic organic chemicals, of the EIA Notification 2006; Category B for a site in a notified industrial area |
| Pollution board consent | Consent to establish and operate as a red category industry, with handling of solvent residue and evaporator salts as hazardous waste |
| Factory and fire safety | Factory licence under the Factories Act and a fire NOC covering solvent storage |
| Business registration | Udyam (MSME) and GST registration |
Checked against official sources in October 2026. Rules vary by state; confirm with the State Drugs Controller and pollution board before you apply.
Market for APIs in India
Demand comes from the formulation units that turn APIs into tablets, capsules and syrups, and from export buyers. Government policy is pushing domestic supply of bulk drugs that India has relied on imports for.
The PLI scheme for 41 bulk drugs selected 48 projects; by December 2024, 34 were commissioned, covering 25 products. Up to March 2025, PLI plants had sold ₹1,817 crore of output, including ₹455 crore of exports.
- Tablet, capsule and syrup formulation units
- Contract and loan-licence manufacturers
- Larger pharmaceutical companies seeking a second source
- API traders and merchant exporters
- Overseas formulation companies
Where a new unit wins: formulators rarely switch API suppliers on price alone. A unit that passes their audit and delivers a consistent impurity profile batch after batch tends to stay on the vendor list.
Mistakes that weaken an API project report
- One price for every molecule
API prices vary many times over between molecules. Revenue should come from the planned product mix and campaign days, not a single average picked from the market.
- Assuming full solvent recovery
Every distillation loses some solvent. Make-up solvent is a fifth of raw material cost in our sample; leaving it out overstates profit.
- Sales from the first month
Formulation buyers audit the plant and test validation batches before placing regular orders. Working capital and the loan moratorium should cover that gap.
- Under-budgeting effluent treatment
High-salt, high-COD effluent needs an evaporator, and the clearance conditions may ask for more. A cheap ETP line in the cost sheet is a common reason for later overruns.

Active Pharma Ingredients API project report: frequently asked questions
What is the active pharma ingredients API manufacturing plant cost in India?
Cost depends on how many molecules you make, the reactor volume and how far effluent is treated on site. Our sample report models a 250 kg per day multi-product unit at ₹650 lakh, including ₹339 lakh of plant and machinery. Units with larger reactor trains, zero liquid discharge or export-grade facilities cost several times more. Your project report prices your own molecules, site and approvals.
What is the difference between an API plant and a formulation plant?
An API plant makes the active drug substance through chemical reactions, purification and drying, and sells it as powder in drums. A formulation plant buys that API and turns it into tablets, capsules or syrups. API plants need reactors, solvent recovery and heavier effluent treatment, and they fall in the red pollution category, while formulation units are mostly mixing and packing.
Which licence is needed to manufacture APIs in India?
You apply in Form 24 for a drug manufacturing licence in Form 25 from the State Licensing Authority, for drugs outside Schedules C, C(1) and X. The premises are inspected first, and the licence names your approved technical staff. The plant must also meet revised Schedule M good manufacturing practices, notified on 28 December 2023.
Does an API plant need environmental clearance?
Yes. Bulk drug manufacture falls under item 5(f), synthetic organic chemicals, of the EIA Notification 2006. Projects inside a notified industrial area are appraised by the state authority as Category B. You also need consent to establish and operate from the State Pollution Control Board, because pharmaceutical manufacturing is in the red category.
Is API manufacturing profitable?
In our illustrative sample, the 250 kg per day unit earns a net profit of ₹182 lakh on sales of ₹1,115 lakh in Year 3. That is a 28% return on investment, with break-even at about 33% of capacity. Real results depend on the molecules chosen, yield per batch, solvent recovery and how fast buyers approve you as a supplier.
Are there government incentives for API manufacturing?
The PLI scheme for bulk drugs, with an outlay of ₹6,940 crore, supports new plants for 41 identified key starting materials, intermediates and APIs. Application windows are limited, so check the Department of Pharmaceuticals for current calls. The bulk drug parks in Gujarat, Himachal Pradesh and Andhra Pradesh are building shared effluent treatment, solvent recovery and steam facilities.
Is the sample report free and can I use it for a loan?
The sample is free to view and download. It is an illustrative report that shows the format and depth of an NPCS report. Its figures are representative and not based on a real project, so it should not be submitted to a bank. For a loan, NPCS prepares a report for your own molecules, capacity, site and lender.
